Bike not starting could it be the check seat valve

I have rebuilt my carby with an all balls kit, a new original yamaha  diaphragm and o rings for the fuel pump and a check seat valve I don't know if the check seat valve is new so is there meant to be a noticeable hole in the check valve because I can't see one, I can prime the throttle and use the choke and it will fire up but won't start, I put back the original pilot and main but it's still the same, I have put the carb body in an ultrasonic cleaner, I see a stream of fuel from the air filter side when I use the throttle so the fuel pump is OK and my bike was running OK with a copy carby of ebay, I put the needle clip in the middle and the air/fuel screw at 2 turns out i even tried 1 1/2 I have never used this original carby before I got it with the bike. Do you guys think it has a blocked valley somewhere the only thing this carby is missing is the outside rubber boot on the choke I tried swapping the choke copy over but it's 0.05 mm to big 

 

The all balls kit has a 50 pilot and a 137 main, the original has a 50 pilot and a 142 main

I meant to say the check valve from the float bowl the small brass piece under the rubber gasket not the check seat valve also how do you clean the small brass jet in the float bowl the one under the rubber gasket
